As you have a large enclosure and multiple hamsters I'd ditch the idea of getting a 4kg and get a 10kg. I only remove soiled bits from my cages. I only take it all out and properly sift through it once a month and the majority of the bedding goes back.

I have roughly 2500sq inches of floor space for my hams in total. Out of a 10kg bag I've used 5kg roughly that's filled up approximately 1000sq inches of space at a depth of 4" approx. But that was topping up the cages not starting from scratch. Also depends on how deep you want the bedding and if you use any other substrate. For example all mine have a sand pit and a 2 inch layer of aspen or paper pellets on the bottom.
